| 内容記述 | It has been reported that sea firefly Vargula hilgendorfii encodes at least two copies of the luciferase gene, each consisting of 13 exons. PCR amplification and agarose gel electrophoresis of the region from exon 5 to exon 6 of the gene revealed four DNA fragments, suggesting that V. hilgendorfii has two additional copies. To determine nucleotide sequences of these fragments, the fragments in the agarose gel were individually poked with the tip of a toothpick, immersed in the PCR reaction solution, and amplified again with the same primer set, and subjected to direct sequencing. The results showed that the fragments contained sequences homologous to exon 5 and exon 6 of the luciferase gene, indicating that V. hilgendorfii has at least four copies of this region. The PCR-based sequencing method used in this study is extremely simple compared to the standard method involving genetic recombination experiments, and is applicable to experiential learning for high school students. | |||||||||||||
