{"created":"2023-05-15T12:04:23.719101+00:00","id":8937,"links":{},"metadata":{"_buckets":{"deposit":"ce836012-6f58-4c64-9361-d8b5e0aeda4c"},"_deposit":{"created_by":3,"id":"8937","owners":[3],"pid":{"revision_id":0,"type":"depid","value":"8937"},"status":"published"},"_oai":{"id":"oai:iwate-u.repo.nii.ac.jp:00008937","sets":["1504:1505"]},"author_link":["70916","70915"],"item_28_alternative_title_23":{"attribute_name":"その他のタイトル","attribute_value_mlt":[{"subitem_alternative_title":"Characteristics of nutrient absorption in growing calves"}]},"item_28_biblio_info_7":{"attribute_name":"書誌情報","attribute_value_mlt":[{"bibliographicIssueDates":{"bibliographicIssueDate":"2000-03-01","bibliographicIssueDateType":"Issued"},"bibliographic_titles":[{}]}]},"item_28_date_6":{"attribute_name":"登録日","attribute_value_mlt":[{"subitem_date_issued_datetime":"2011-10-04"}]},"item_28_description_11":{"attribute_name":"抄録","attribute_value_mlt":[{"subitem_description":"科学研究費補助金[基盤研究C](課題番号10660264)研究成果報告書","subitem_description_type":"Abstract"}]},"item_28_description_12":{"attribute_name":"Abstract","attribute_value_mlt":[{"subitem_description":"反芻動物の消化吸収能は、ルーメンの発達に伴い大きく変化する。しかし、栄養生理的には不明な点が多々残されている。そこで、子ウシの成長、発育に伴う栄養素吸収特性を調べ、その調節の可能性について、特にグルコース吸収能に焦点を当て、SGLT-1を指標として検討した。\n\n実験に先立ち、子ウシの腸管粘膜上皮細胞に存在するSGLT-1・mRNA定量法の検討を行った。次いで子ウシ(計27頭)を供試し、腸管粘膜細胞からtotal RNAを抽出し、RT-PCRによりサブクロ―ニングしたSGLT-1・のcDNAをプロ―ブとしてノーザンブロッティングを行った。離乳前、後および十二指腸から回腸上部の間で比較した。その結果、離乳前の子ウシでは、十二指腸でSGLT-1・mRNAの発現が弱く、幽門部より遠位になるに従い徐々に発現が強くなった。一方、離乳後では十二指腸から回腸上部にかけてほぼ同様のSGLT-1・mRNAの発現が見られた。しかし、この発現には個体による変動も大きい可能性が示唆された。子ウシの場合、消化吸収能の発育には12週齢ではまだ充分ではなく、もう少し成育の進んだ状態まで検討する必要があるものと思われた。\n\n次に、子ウシの離乳前後におけるα-グルコシダーゼおよびβ-ガラクトシダーゼ活性についても検討した。α-グルコシダーゼ活性は、離乳前の子ウシの方が低く、部位による顕著な違いも見られなかった。一方、離乳後では、離乳前に比べておよそ2倍の活性が認められた。また、消化管を部位別に比較すると、十二指腸に比べ下部になるに従い高い活性を示した。β-ガラクトシダーゼ活性については、離乳前、離乳後ともに大きな違いは見られなかった。\n\n以上より、子ウシの場合、12週齢では、まだ消化吸収能は発育途上にあり、更に週齢の進んだ動物を供試して検討する必要があるが、グルコース吸収能を調節する可能性は充分にあると考えられた。","subitem_description_type":"Other"},{"subitem_description":"In the small intestine, the brush-border NaィイD1+ィエD1/glucose cotransporters (SGLT-1) play a major role in the absorption of glucose . The objectives of this experiment were to investigate the developmental changes of the intestinal function for glucose absorption and to recognize the assay for SGLT-1 in new born calves. The SGLT-1mRNA was determined by Northern blot analysis . The mucosal SGLT-1mRNA levels were lower in duodenum and gradually increased in proximal intestine before weaning calves. After normal weaning at 5 weeks old , the expression of SGLT-1 in the small intestine was not different between the location of intestine.
