{"created":"2023-05-15T12:04:24.192712+00:00","id":8946,"links":{},"metadata":{"_buckets":{"deposit":"2344736a-a6c2-43c4-b44b-a405f1a1a2bb"},"_deposit":{"created_by":3,"id":"8946","owners":[3],"pid":{"revision_id":0,"type":"depid","value":"8946"},"status":"published"},"_oai":{"id":"oai:iwate-u.repo.nii.ac.jp:00008946","sets":["1504:1505"]},"author_link":["70933","70934"],"item_28_alternative_title_23":{"attribute_name":"その他のタイトル","attribute_value_mlt":[{"subitem_alternative_title":"The enzymatic mechanisms in accumulation of AGEs and its regulation by flavonoids"}]},"item_28_biblio_info_7":{"attribute_name":"書誌情報","attribute_value_mlt":[{"bibliographicIssueDates":{"bibliographicIssueDate":"2005-06-01","bibliographicIssueDateType":"Issued"},"bibliographic_titles":[{}]}]},"item_28_date_6":{"attribute_name":"登録日","attribute_value_mlt":[{"subitem_date_issued_datetime":"2011-12-16"}]},"item_28_description_11":{"attribute_name":"抄録","attribute_value_mlt":[{"subitem_description":"科学研究費補助金[基盤研究C](課題番号14560090)研究成果報告書","subitem_description_type":"Abstract"},{"subitem_description":"巻末の学術雑誌掲載論文は削除しております.","subitem_description_type":"Abstract"}]},"item_28_description_12":{"attribute_name":"Abstract","attribute_value_mlt":[{"subitem_description":"非酵素的糖化反応(グリケーション)に対してフラボノイドが阻害効果を持つかどうか、ルチンを用いて検討した。糖尿病ラット筋肉、腎臓、血清タンパク質のグリケーション前期段階の指標となるフルクトースリジン含量は顕著に増加したが、0.2%ルチン食の摂食で有意に抑制された。さらに後期段階生成物(AGE)は腎臓や血清で顕著に抑制された。これらの結果から、フラボノイドは抗酸化だけではなく、グリケーションの多くのステップで阻害していることが示唆された。\n\n糖尿病ラットにおいてはきわめて反応性の高いα-ジカルボニル化合物量が増加した。α-ジカルボニル化合物の生成の律速酵素であるアルドース還元酵素活性は、多くの組織で糖尿病により増加したが、ルチンの摂取で減少した。また、ルチン以外にも、カテキンやアントシアニンを豊富に含むヤマブドウ抽出物でもアルドース還元酵素活性が抑制された。しかし、カテキンの阻害作用は弱かった。α-ジカルボニル化合物の分解に関わるグリオキザラーゼ活性は、糖尿病ラットで低下した。このことから、糖尿病ラットではα-ジカルボニル化合物が蓄積されやすい状況にあることが明らかになった。グリオキザラーゼ活性はin vitroではエピガロカテキンガレートやルチンでその活性が増加することを示したが、in vivoでは効果が今回の濃度では認められなかった。\n\n本研究において、プロテアソームはグリケーションに対して他のタンパク質より抵抗性があることが明らかになった。しかし、酸化修飾タンパク質とは異なり、AGE化されたタンパク質に対しては、ユビキチン化がされているものの、分解は増加しなかった。これは高度に架橋された場合にはもはや分解できず、蓄積されてしまうことを示している6以上のように、AGEの生成には酵素的な作用が重要であり、フラボノイドによりその一部が好ましい方向に制御できる可能性が示唆された。","subitem_description_type":"Other"},{"subitem_description":"We investigated that flabonoids shows inhibition of glycation using rut in and flavonoids rich extract from wild grape. N_ε-fructoselysine contents of skeletal muscle, kidney and serum proteins markedly increased in streptozotocin-induced diabetic rats. 0.2% rutin supplemented diet suppressed these increases. Furthermore, the increased AGE contents in diabetic rats were also inhibited by dietary rutin. Dietary wild grape extract suppressed AGE content in kidney and serum protein in the same extent of dietary rutin. These results suggested that flavonoids have effects of inhibition in many steps in glycation reaction.\n\nThe increased α-dicarbonyl compounds content in liver of streptozotocin-induced diabetic rats was decreased by feeding of rutin and wild grape extract supplemented diet. The increased activity in aldose reductase, the rate-limiting enzyme in the production of α-dicarbonyl compounds in diabetic rats was attenuated by feeding of these flavonoids supplemented diet. In vitro experiment showed that rutin and quercetin inhibited the activity of aldose reductase. On the other hand, the activity of glyoxalase, a decomposing enzyme against α-dicarbonyl compounds reduced in diabetic rats. The activity of glyoxalase was inhibited by wild grape extract in vitro, but not in vivo.\n\nProteasome activity itself was more resistant against glycation than other enzymes. Oxidatively modified proteins are easily degraded by proteasome. Although AGE proteins were ubiquitinated, degradation of these proteins were not increased.
