Cspp.）の育種を進める上で必要となる知見を得るため、組織培養技術の開発ならびに遺伝資源として重要なリンドウ及び関連植物群の系統解析について、以下の内容について研究を行った。\n１．リンドウ科園芸植物の再分化系の確立\n　リンドウの再分化系を確立することを目的として、葉片からのカルス形成およびシュート誘導における植物成長調節物質の影響について検討した。G. scabraおよびG. trifloraでは、2,4-DとBAでカルスを誘導でき、さらに、G. scabraではBAおよびTDZで形成されたカルスから不定芽と不定胚を誘導できた。また、TDZを用い葉片培養を行い、G. scabraでは不定胚を、G. trifloraでは不定芽を誘導でき、リンドウの組織培養ではTDZが効果的であることが明らかとなった。G. scabraで誘導された不定胚について組織学的観察を行い、不定胚は、形成カルス内および葉片の表皮細胞から由来していることが明らかとなった。次に、エキザカム(Exacum affine)の再分化系の検討を行い、葉片培養では2,4-DとBAでカルス形成し、さらにBAによりカルスからの不定芽形成が可能となった。また、無菌植物体の根に形成したカルスから再分化がみられ、再分化能のある外植片として利用可能なことを明らかにした。\n２．リンドウ属植物のプロトプラスト培養\n　リンドウ属３種６系統及びトルコギキョウ、エキザカムからのプロトプラストの単離方法を検討した。３種類の酵素組成を用い、すべての植物でプロトプラストが得られたが、種間差や系統間差が認められた。次に、G. scabraおよびG. trifloraのプロトプラスト培養について培養条件を検討した。G. scabraのプロトプラスト培養では、MS培地の硝酸アンモニウム濃度を検討し、1/4の濃度に改変したNAA 2mg/lとBA 1mg/lを含む培地でコロニー形成し、カルス形成と根の分化がみられたが、植物体再生に至らなかった。また、プロトプラストの褐変化防止物質の効果を調査したところ、それらの効果はみられなかった。一方、G. triflora,では、BAよりもTDZがプロトプラストの分裂およびコロニー形成に効果的であったが、根の分化がみられただけで植物体再生には至らなかった。\n３．分子生物学的手法によるリンドウ属植物の系統解析\n　G．trifloraとG. scabraの種内および種間の変異性を明らかにするため、15系統を用いRAPD法により解析した。８個のプライマーで53の多型バンドが得られ、このデータを基に数量化理論III類及びクラスター分析を行った結果、両種は異なった群に分かれた。両種を識別するためのSCARマーカーを作成し、ある程度種を識別できるマーカーとして利用できることが明らかとなった。次に、葉緑体DNAのPS-IDの塩基配列に基づきリンドウ属17種およびその他の属(Eustoma, Exacum, Swertia)３種の類縁関係を調査した。その結果、リンドウ属とその他の属に大きく２つに分かれ、リンドウ属内の種については、2n＝26の染色体数を持つ種は、すべて同一のPS-ID配列を持っており近縁と考えられたが、2n＝36など染色体数の異なる種は部分的に異なる配列を示し系統的に異なると考えられた。特に、G.
